High Performance Liquid Chromatography (HPLC)

Expert HPLC Services: Chemial Analysis at FAI

FAI utilizes two HP 1090 High Performance Liquid Chromatography (HPLC) systems for our identification of compounds. Also called a "High Pressure Liquid Chromatograph," the HPLC is used in many applications including drugs in biological samples, pharmaceutical analysis, food and nutritional analysis, surfactant analysis, analysis of ions, polymer analysis, ionic compounds and organic compounds.

In an HPLC, the sample is introduced into a mobile phase (solvent) and pumped at high pressure through a column. The column consists of phase-coated chromatographic packing material, such as beads, that acts as a stationary phase. The column materials and mobile phases used are chosen depending on what chemicals are to be analyzed and what matrix they are in. As the mobile phase moves through the column, the material is separated into its different components. These components elute from the HPLC at different times called "retention times." Retention times are dependent on the interaction between the molecules in the sample and the solvent used in the system. Different retention times allow for an identification of what is present in the material.

FAI uses Refractive Index, Diode Array - UV Spectral Detection (DAD), Conductivity Detection, and Amperometry Detection. RI detection can be used when the chemicals of interest do not have any strong UV chromaphores and UV-DAD can not be used. The DAD simultaneously monitors UV and visible light transmission through the sample. These spectra are matched to the retention times in the chromatogram, allowing detailed analysis of sample components by multiple techniques.
